Callum and Paul Smith, Gavin McDonnell and Kid Galahad have been confirmed on the undercard of Kell Brook's showdown with Gennady Golovkin on September 10.
IBF welterweight champion Brook (36-0-KO25) will step up two weight divisions when he challenges WBA Super, WBC and IBF middleweight king Golovkin (35-0-KO32) at The O2, live on Sky Sports Box Office.
Liverpool's super-middleweight brothers Callum and Paul will also be in action at the sold-out arena, where unbeaten star Callum will put his WBC silver title on the line against tough Hungarian Norbert Nemesapati while big brother Paul will continue his bid to secure a third world title shot.
McDonnell is also hoping to secure a world title shot and the unbeaten Doncaster super-bantamweight will defend his WBC silver belt for the first time.
Brook's gym-mate Galahad, also a super-bantam, returns to the ring after stopping Simas Volosinas in April in the second fight of his comeback.
The support card also features Lee Haskins defending his IBF bantamweight title against bitter rival Stuart Hall, who he beat to win the European strap in 2012, and Charlie Edwards challenges IBF flyweight champion Johnriel Casimero in just his ninth fight as a professional.
Martin J. Ward and Andy Townend meet for the vacant British super-featherweight title, Conor Benn boxes for the fourth time in the paid ranks and middleweight Craig Richards attempts to stretch his unbeaten record to five.
"The card is now set for the biggest night of boxing in 2016," said promoter Eddie Hearn.
"Callum Smith has one more hurdle before the mandatory against WBC champ Badou Jack is called on September 13 and we are looking for him to make a statement on this huge platform.
"Gavin McDonnell is sitting pretty for a shot at the world title later this year and Paul Smith is also in the hunt with one eye on the winner of the rematch between Giovanni De Carolis and Tyron Zeuge for the WBA strap.
"With the three world title fights and a Lonsdale Belt clash, this night is packed with action as we count down to the moment of truth between unbeaten divisional number ones Gennady Golovkin and Kell Brook."
