The Titans opted to bowl first, having left out Dale Steyn and Morne Morkel, and immediately made a breakthrough as Albie Morkel bowled Wayne Parnell with the fifth ball of the innings.
Colin Ackermann (48) and Colin Ingram (41) shared a 63-run third-wicket partnership but spinner Shamsi caused havoc with the lower order batsmen as the Warriors were bowled out for 143 in 18.1 overs.
The home side's response got off to a poor start as Quinton de Kock fell early when JJ Smuts took a brilliant catch at mid-wicket to dismiss the wicket-keeper for just one off Andrew Birch.
De Villiers came to the crease when Parnell has Heinrich Klassen (24) caught and the South African Test captain shared a century partnership with Markram (56 not out) to seal a convincing win.
