Holders St Helens are boosted by the return of captain Paul Sculthorpe for their trip to face Hull KR at Craven Park, where they lost in the opening round of Super League.
Sculthorpe has recovered from a pectoral injury while second rower Paul Clough returns in place of Mike Bennett, who sustained a potential career-threatening shoulder injury in Sunday's win at Hull.
Saints also welcome back leading tryscorer Ade Gardner, who was forced to sit out the last game after sustaining burns on his arm while opening the radiator cap of his car.
St Helens' Great Britain forward Maurie Fa'asavalu, who missed out on his side's cup final win over Wigan in 2004, is aiming to complete a hat-trick of triumphs and has no doubt the Challenge Cup beats the Super League Grand Final in terms of prestige.
"It's the best cup in rugby league over here," he said. "Super League only started in 1996 but the Challenge Cup has got history and all the teams, even from lower divisions, play in it.
"We've won it three times since I've been here and, although I missed 2004, the experience of sitting there made me say to myself I've got to make sure next time I'm playing instead of watching. It's a great experience."
