Challenge Cup holders Warrington ended Featherstone's nine-match winning run, with Richie Myler scoring a hat-trick in a nine-try display from the Wolves.
But Rovers battled bravely throughout, clawing back to within four points after trailing 16-0 in the first eight minutes.
Featherstone had won all six matches in the Co-operative Championship, two in the Northern Rail Cup and one in the Challenge Cup third-round.
But the Wolves, who hammered Featherstone 56-8 in the fifth round last season, took only two minutes to go ahead with Simon Grix going over in the corner and Ben Westwood landing the touchline conversion.
Then Grix ploughed through Jessie Joe Parker's tackle for Chris Riley to bag his 14th try of the season to make it 10-0.
Featherstone looked to have no answer to Warrington's power and pace with Adrian Morley's slipped pass to Paul Wood allowing man-of-the-match Myler to dart 35 metres to score before a spirited fightback.
Rovers did manage to open their account after 15 minutes with Liam Finn's well judged kick causing problems on the Wolves left flank and Papua New Guinea international Parker touched down with Kyle Briggs adding the conversion.
Warrington were then rocked with Parker outjumping the home side's casual defence to gather Briggs' high kick and notch his second try to cut the deficit to 16-12.
A Ross Divorty break caused problems for Warrington with Parker just failing to complete his hat-trick from Ian Hardman's kick.
Power over
Warrington went 24 minutes without scoring and some nerves were showing, before substitute Vinnie Anderson took Chris Bridge's pass to power over and then David Solomona supported Richie Mathers to add a fifth try and extend the lead to 26-12.
Bridge carved out a second try for Myler to stretch the advantage to 32-12 at the interval.
Featherstone did well to halt Matt King over the line but could not stop Solomona making it 36-12 after 52 minutes.
Myler completed his hat-trick with a scorching 50-metre try before Williams saved a touchdown with a superb tackle on Liam Welham.
But Zak Hardaker went over for Rovers' third try 10 minutes from time and then Stuart Dickens crossed for a score his efforts deserved.
Riley's second try in the last minute from Myler's kick completed the scoring, with Bridge adding his second conversion.
