Winger Nick Cummins scored two of the Wallabies' seven tries to make sure they are still yet to suffer a defeat against their opponents in 16 meetings between the two countries.
The Azzurri actually led 10-0 at one stage after an early try from Australian-born full-back Luke McLean but were punished time and time again for their errors.
Alberto Di Bernardo kicked over a penalty in the fourth minute and then also added the extras after McLean finished off a counter-attacking move down the right-hand side.
The try seemed to kick Australia into life, captain Ben Mowen touched down on 15 minutes and Quade Cooper made up for an earlier penalty miss with the conversion.
On target
The fly-half was also on target after Tevita Kuridrani went over on 21 minutes, though he couldn't add the extras after Cummins' first try of the afternoon on the half-hour mark following excellent work by scrum-half Will Genia
Still, the visitors led 19-10 at the break and quickly set about extending their advantage at the start of the second half.
Adam Ashley-Cooper offloaded for Cummins to complete his brace and then went from provider to scorer, driving over to the side of the uprights to leave Cooper with an easy kick.
Italy were given brief hope of a fightback when replacement Lorenzo Cittadini bundled the ball over shortly after to add five points, though Alberto Di Bernardo missed the kick.
But Australia were in no mood to allow their opponents to rally, Joe Tomane and Israel Folau touching down for a quick-fire brace that saw replacement Christian Lealifano successfully convert on both occasions.
The home team did at least have the last word after leaking 50 points, second-half sub Tommaso Allan, formerly a Scotland player at junior level, registering his maiden Italy try in the final minute.
