Both teams are coming off wins in Round One after La Rochelle smashed Rovigo on the road and Agen upset Gloucester at home.
La Rochelle have opted to rotate their squad with Seru Rabeni coming into the side as one of 10 changes.
Agen, themselves, have made 14 changes - with only full-back Florent Cazeaux retaining his place.
