Poole, 26, has been sidelined since October 2011 after rapturing knee ligaments against Treviso and then suffering a further setback in pre-season.
Influential lock Andrew Coombs also returns from the treatment and takes his place in the Dragons starting XV on Saturday having recovered from a knee injury of his own.
In total Dragons coach Darren Edwards makes 11 changes to the side that was well beaten by Treviso in the RaboDirect PRO12 last time out.
Coombs forms part of a whole new front five in the pack, while in the three-quarters youngster Jack Dixon partners the experienced Andy Tuilagi in the centres.
Mogliano will be looking to keep things competitive after suffering heavy defeats in their first two outings in the competition.
A 71-7 thrashing on their debut appearance in the event against Bayonne was followed by a 59-12 hammering on home soil by Wasps and the Italian outfit will be hoping Saturday's clash is more of a contest.
