Top-seeded David Ferrer suffered a surprise defeat to Marcos Baghdatis at the ATP AEGON Open in Nottingham on Tuesday while Britain's Aljaz Bedene and James Ward both suffered defeats.
The world No 7 lost his first two service games to Cypriot Baghdatis, who earned his first win over a top-10 player in more than two years with a 6-2 7-6 (7-4) win.
"For me, the first set was difficult because it was the first set back on grass," Ferrer said after his first appearance in Nottingham in 11 years.
"He (Baghdatis) played more hours on court, and had more confidence in his game."
He will face German teenager Alexander Zverev, who earned a notable win of his own as he beat 13th seed Thomaz Bellucci 6-5 (5-7) 6-3 6-4.
Britain's Aljaz Bedene was beaten 7-6 (7-4) 6-4 by eighth seed Adrian Mannarino, the Frenchman setting up a clash with Italy's Simone Bolelli who beat 10th seed Martin Klizan 6-3 6-7 (4-7) 6-1.
There was double duty for Tim Smyczek who came from a set down to beat Britain's James Ward 1-6 7-6 (7-3) 6-1 but running out of steam in a 6-1 6-3 loss to 16th-seeded Czech Jiri Vesely.
Second seed Gilles Simon needed three sets, two tie-breaks and more than three hours to see off Spain's Marcel Granollers.
Simon saved three match points when Granollers led 6-4 5-3 40-0 and recovered to win 4-6 7-6 (7-3) 7-6 (7-0).
Queen's Club semi-finalist Viktor Troicki, seeded sixth, lost 6-4 6-2 to big-serving Sergiy Stakhovsky, who made his name after knocking Roger Federer out of Wimbledon two years ago.
There were no such problems for Feliciano Lopez, Leonardo Mayer and Pablo Cuevas, seeded three, four and five respectively.
Dominic Thiem, Adrian Mannarino, Sam Querrey and Jiri Vesely all advanced safely.
