Jake Ball and Justin Tipuric have been released from the Wales squad for Saturday's clash with South Africa because of injury.
Scarlets lock Ball faces an extended spell on the sidelines as he needs surgery on a dislocated shoulder, while Tipuric has returned to the Ospreys for treatment on a quad issue.
Tipuric had been absent for a month with a similar injury but returned to action as a second-half substitute in the 33-18 defeat at the hands of New Zealand last Saturday.
Rhys Webb remains with Warren Gatland's squad despite having suffered a head injury against the All Blacks - he is going through the sport's concussion protocols.
Scott Williams is also a doubt with an ankle injury suffered in the bruising game at the Principality Stadium.
Wales are looking to bounce back from defeats against the other two southern hemisphere giants when they take on the Springboks, with their only autumn win so far having been a narrow one against Georgia.
