Premier League sides Aston Villa and West Brom squeezed into the fourth round of the FA Cup but non-league Eastleigh's dream is over.
The lowest-ranked side left in the competition took an early lead against Bolton through Joe Partington (11) and then came from behind after Gary Madine (39) and Dean Moxey (43) had wrestled back momentum for the hosts.
Eastleigh equalised but Darren Pratley - the man who forced the replay - struck (58) to settle a five-goal thriller and earn the Championship side a fourth-round tie with Leeds.
Premier League strugglers Aston Villa were forced to go again after being held to a 1-1 draw at Wycombe but had to wait until 15 minutes from time to seal a date with Manchester City, Ciaran Clark finally breaking the deadlock (75) before Idrissa Gana (90) saw off the League Two side.
West Brom also only narrowly edged through, Salomon Rondon's first FA Cup goal ousting Bristol City at Ashton Gate.
Rondon settled the tie 1-0 in Albion's favour with only his fourth goal for the Baggies since completing a £12m move from Zenit St Petersburg last August.
Gary Roberts put Pompey ahead from the spot before Marc McNulty extended the hosts lead, Arsenal loanee Ainsley Maitland-Niles' (60) reply in vain as Paul Cook's men secured a plum home draw against top-flight neighbours Bournemouth.
More than 7,000 Cobblers fans made the trip to Stadium:mk, but penalties from Ben Reeves and Simon Church either side of Josh Murphy's strike eased the Championship side to victory
Elsewhere, Reading booked a home clash with Walsall after winning a topsy-turvy tie 5-2 against Huddersfield.
The Terriers had gone 2-0 up before seeing Jonathan Hogg sent off and a subsequent Matej Vydra hat-trick helped the hosts seal a home date with Walsall.
Yeovil paid the price against Carlisle, the visitors afforded extra-time after Francois Zoko missed a last-gasp spot-kick, and then sealing a 5-4 win on penalties themselves to earn a tie with Everton.
Bradford and Bury played out a goalless draw across 120 minutes but the Lancashire side made it through after a 4-2 spot-kick triumph, Hull their next obstacle.
