Rossi has presided over three losses in a row and, heading into the international break, Samp are third bottom and a point away from safety.
Sunday's 2-1 reverse to Fiorentina was Rossi's last act and Serbia coach Sinisa Mihajlovic is a reported target as his replacement.
"Sampdoria announces that it has relieved Delio Rossi and his staff of their duties," a club statement read on Monday night.
"The company would like to thank everyone for their commitment and professionalism over these past months and wish you the best of luck.
"The management of the first team will be temporarily entrusted to Francesco Pedone."
Rossi was phlegmatic when asked about his future after the Fiorentina defeat.
"If there is a time where you don't feel that the trust is there, you shouldn't wait to see how things unfold," he said.
"I am a man of the world and I think that if the confidence is there, then it's normal that one continues, but if it isn't, then it is better to end the rapport regardless of the results."
The well-travelled 52-year-old only joined Samp in December last year for what was his 16th job.
