The Uruguay international, who is on a season-long loan at Levante from Valencia, sustained the injury during their goalless Primera Liga draw at home to Real Madrid on Saturday.
Medical examinations have revealed ruptured ligaments in the 28-year-old's right knee, and doctors have predicted a half-year recuperation period.
His injury will strike a severe blow for Levante, who sit third bottom of the La Liga table having collected just four points from five league matches.
