Spurs signed the highly-rated defender from the Blades last summer before immediately allowing him to return to Bramall Lane on a season-long loan deal.
The Premier League club have been monitoring his progress closely and decided just before Monday's transfer deadline to recall him.
Sheffield United officials tried to dissuade Tottenham but were unsuccessful and they must now cope without Walker.
Blades manager Kevin Blackwell still has options at right-back for the Championship clash with Watford on Tuesday.
Derek Geary could replace Walker or Nyron Nosworthy might come in for his debut after arriving on loan from Sunderland.
Walker, meanwhile, will be determined to make an impact at White Hart Lane after gaining further experience with Sheffield United over the past few months.
