Aston Villa host Notts County, Newcastle are at home to Northampton Town, Sunderland entertain Exeter City, Swansea host York City while Bournemouth travel to Hartlepool United.
Leicester, who suffered a shock second round exit last season, are one of a further six top-tier sides in action as they face League One's Bury.
Crystal Palace, West Bromwich Albion and Everton all face League One opposition in Shrewsbury Town, Port Vale and Barnsley respectively, while Watford travel to Preston North End and Norwich to Rotherham in ties between Premier League and Championship sides.
Elsewhere, Portsmouth, who beat Derby County in Round One, host Reading and Hull City and Queens' Park Rangers are at home against Rochdale and Carlisle United respectively.
Early season Championship leaders Ipswich Town are away to Doncaster Rovers, while Gillingham, unbeaten in League One, travel to Birmingham City.
Everton's trip to Oakwell will be live on Sky Sports 1 on Wednesday from 7:30pm.
