That sent Samp down with already-doomed Bari and Brescia and means next season the Blucerchiati will play in Serie B for the first time since 2002/03.
Samp had won just one of their previous 12 and fell behind to a Fabrizio Miccoli strike in first-half stoppage time.
Jonathan Biabiany's 49th-minute header drew the hosts level but Jeda then gave Lecce a 51st-minute lead at Bari, meaning Samp would have to win to keep their survival hopes alive heading into the final day.
However, an Andrea Masiello own goal in the 80th minute secured victory for Lecce and an 85th-minute winner by Palermo sub Mauricio Pinilla sealed Samp's fate.
Udinese climbed back above Lazio into the final Champions League spot thanks to a 2-0 win at
Chievo.
Mauricio Isla and Kojo Asamoah scored midway through each half to send Francesco Guidolin's side back into fourth place.
Napoli earned the point they needed to secure a third-placed finish and a passage straight through to the Champions League group stage as they drew 1-1 with second-placed
Inter Milan.
Defeat would have meant Udinese could still catch Napoli on goal difference heading into the last weekend but defender Juan Camilo Zuniga cancelled out Samuel Eto'o's opener for Inter, who have already lost the title race to rivals AC Milan.

Cesena began the day four points above the drop zone and clinched survival with a 1-0 home win against
Brescia, Emanuele Giaccherni scoring the game's only goal in the 59th minute.
Cesena therefore edged up to 15th place above
Bologna, who drew 1-1 at ninth-placed
Fiorentina after Alessio Cerci's 19th-minute opener for the hosts was cancelled out by Gaston Ramirez.
The Uruguayan midfielder struck just two minutes after coming on in the 47th minute, with Fiorentina defender Per Kroldrup sent off in the 53rd minute before Ramirez followed after two bookings late on.
