Reigning Eredivisie champions Twente were the dominant team in the first half and scored the decisive goal of the match after just eight minutes.
Ajax keeper Maarten Stekelenburg played a poor pass to full-back Gregory van der Wiel which was seized upon by De Jong who slotted home from a narrow angle.
Twente pressed for a second goal and saw Theo Janssen hit the crossbar before Stekelenburg beat away De Jong's header.
Ajax were reduced to 10 men before half-time when Suarez, who was famously sent off during Uruguay's World Cup quarter-final victory over Ghana, saw red for a rash challenge.
The second half offered little in the way of goalscoring opportunities as new Twente coach Michel Preud'homme ensured his reign started on a winning note.
Twente will begin the defence of their Dutch title on Friday against Roda JC Kerkrade, while Ajax face Groningen next Sunday.
