After a goalless first half, Groningen were ahead just minutes into the second period as Andreas Granqvist scored from the penalty spot.
Slovenian star Tim Matavz doubled their advantage on the hour, and just minutes later Petter Andersen grabbed a third.
Marcus Pedersen pulled one back for Vitesse on 79 minutes, before Tim Sparv added a fourth for the home side.
The win takes Groningen within a point of joint leaders PSV Eindhoven and FC Twente - but both have played a game less. Vitesse remain just two points above the bottom three.
