The 26-year-old hurt his shoulder, whilst playing in goal during a Brazilian training session at the Ulsan Mansu Stadium.
The Brazilians were preparing for their opening game with Turkey on Monday.
The Samba side's team doctor Jose Luis Runco confirmed that the Roma man would miss the whole of the tournament.
"The shoulder is dislocated," said Runco.
"This injury takes four weeks to recover so he will not be able to play any more in this World Cup."
Runco also revealed to the media that Brazil had made a request to Fifa for a replacement and the governing body should sanction the move as the Brazilians are yet to play their first game.
Fifa rules state that a player can be replaced if he is injured up to 24 hours before their first encounter.
Rivaldo, whose shot indirectly caused the injury, blamed Emerson's inexperience in goal for his injury.
"I shot on goal and I think Emerson had a problem because he is not a goalkeeper," said the Barca star.
"He doesn't know how to fall on the floor, that's why he injured the shoulder."
Emerson's injury will now give a chance to either Vampeta or Gilberto Silva to claim a place in the starting XI.
