Goalkeeper Ben Alnwick, midfielder Liam Lawrence and striker Chris Brown are reported to have appeared in a recording with a fourth person - an unnamed woman.
The trio could find themselves in hot water with The Black Cats should they be found to have breached the club's disciplinary rules.
Sunderland marketing and communications director Lesley Callaghan, speaking on behalf of the board, said: "The behaviour of any individual that adversely affects the image or good name of Sunderland football club is obviously disappointing, and we will be handling this matter internally."
Alnwick has agreed to join Tottenham in January, Lawrence is on loan at Stoke with a view to a permanent move, while Brown remains on Wearside, albeit on the periphery of the first team.
Former Sunderland midfielder Martin Woods, who now plies his trade for Rotherham, is reported also to have been present when the filming took place at a flat.
