Danko Lazovic set the home side on their way with the opener on 25 minutes and former Liverpool man Jan Kromkamp doubled the lead on 56 minutes.
Darryl Roberts pulled a goal back for Sparta in the final minute, but Jason Culina made sure of the win with PSV's third goal in the dying seconds.
Ajax remain in second spot after a 2-0 victory over
Utrecht.
Dutch international Klaas-Jan Huntelaar broke the deadlock on 68 minutes before Sander Keller saw red for the visitors three minutes later.
Ajax made their numerical advantage count 13 minutes from time when Johnny Heitinga found the net.

Sandro Calabro netted a consolation for Venlo nine minutes from time only for Danijel Pranjic to add a fifth for Heerenveen one minute from time.
Roda JC missed the chance to close the gap on the leading pack after going down 2-1 to

Boldizsar Bodor gave Roda the lead on 32 minutes from the penalty spot, but Lasse Schoene drew the home side level on the stroke of half-time when he converted from the spot.
