Southall has also recovered from the groin injury that forced him to sit out the 1-1 draw against the Cherries.
Midfielder Curtis Weston is not eligible to play due to the terms of his loan deal from Leeds, but fellow loanee Leigh Mills has been given permission to feature by his parent club Tottenham.
Otherwise, Mark Stimson has no new injury worries or suspensions to contend with as the Gills look to earn a League One scalp in the opening round of this year's competition.
Colchester missing duo
New signing David Perkins and centre-half Pat Baldwin remain Colchester's only absentees as Geraint Williams looks for a morale-boosting result following Saturday's 4-2 defeat at Hartlepool.
Perkins has three games of the five-match ban he picked up for his red card for Rochdale in the League Two play-off semi-final remaining, while Baldwin is still recovering from a dislocated shoulder.
Steve Gillespie will keep his place in attack after scoring twice on his debut against Pools.
