They opened up with victory over Tottenham two weeks ago but Pasquale Marino's men produced an even better display to seal a 2-1 win in the Russian capital.
The Bianconeri's hero was again Italian international Fabio Quagliarella, who grabbed a brace at the Luzhniki Stadium.
Quagliarella hit the post just 11 minutes in, but just 60 seconds later he opened the scoring as he raced onto Simone Pepe's through ball before firing home.
Spartak responded well to going behind and levelled matters just five minutes later when Clemente Rodriguez curled the ball home from just inside the area.
But Udinese had the final word as Quagliarella slotted home from the penalty spot on the hour after he was brought down by Rodriguez.
Udinese also finished the game with ten-man after Giovani Pasquale was handed a second yellow-card with just four minutes left.
Udinese now sit top of Group D with six points, whilst Spartak are without a point - although this was their first game of the group stages.
