Schalke trailed league leaders Leverkusen by a single point ahead of their home game against a Mainz side with just one point from their past four games.
And Jefferson Farfan broke the deadlock in only the 12th minute after latching onto a pass from Lewis Holtby.
There were no further goals as Mainz, currently eighth in the standings, tried in vain to find an equaliser.
Leverkusen will have the chance to return to the summit when they host Borussia Monchengladbach on Saturday afternoon.
