Barcelona were held 1-1 by lowly Hercules in their Copa del Rey last 32, first-leg match on Wednesday.
Barca's Carles Alena struck a sublime effort in the 58th minute to cancel out David Mainz's close-range opener six minutes earlier, but the holders had to settle for a draw.
With El Clasico on the horizon this weekend, manager Luis Enrique made the expected sweeping changes for the trip to Alicante.
But the Spanish champions' struggles in La Liga followed them to Segunda B side Hercules as a much-changed side made it three domestic games without a win after a disjointed performance.
Real Madrid, playing a leg earlier than everyone else ahead of their Club World Cup campaign, breezed to a 13-2 aggregate win over Cultural Leonesa.
Elsewhere, Joaquin Correa scored a hat-trick as Sevilla thumped fourth-tier SD Formentera 5-1 while Atletico Madrid stormed to a 6-0 victory over Segunda B outfit Guijuelo.
Yannick Carrasco scored twice for Atletico and Saul Niguez, Sime Vrsaljko, Angel Correa and Roberto Nunez also netted.
Wissam Ben Yedder netted two penalties for Sevilla to go with Correa's treble.
Villarreal won 3-0 at Toledo, with an own goal setting the La Liga side on their way before strikes from Cedric Bakambu and Samuel Castillejo.
Cordoba scored two late goals to beat Malaga 2-0 while Sergi Gomez netted for Celta Vigo in their 1-0 win at UCAM Murcia.
Granada were 1-0 winners against Osasuna.
